1. Prepare all reagents, samples and standards as instructed in the manual.
2. Add 50 µl of standard or sample to each well.
3. Add 50 µl of Bioaim mix.
4. Incubate 1 h at RT.
5. Wash and add 100 µl of TMB One-Step Substrate Reagent to each well.
6. Incubate 15 min at RT.
7. Add 50 µl of Stop Solution to each well.
8. Read at 450 nm/570 nm.
Galectin-3 is a protein that in humans is encoded by the LGALS3 gene. It is a member of the lectin family, of which 14 mammalian galectins have been identified. Human and mouse Galectin-3 shares approximately 80% amino acid sequence similarity. Galectin-3 has an affinity for beta-galactosides and exhibits antimicrobial activity against bacteria and fungi. It has been shown to be involved in the following biological processes: cell adhesion, cell activation and chemoattraction, cell growth and differentiation, cell cycle, and apoptosis.
Galectin-3 associates with the primary cilium and modulates renal cyst growth in congenital polycystic kidney disease. A correlation between galectin-3 expression levels and various types of fibrosis has been found. Galectin-3 is upregulated in cases of liver fibrosis, renal fibrosis, and idiopathic pulmonary fibrosis. Elevated levels of galectin-3 have been found to be significantly associated with higher risk of death in both acute decompensated heart failure and chronic heart failure populations.
The entire kit may be stored at -20°C for up to 1 year from the date of shipment. Avoid repeated freeze-thaw cycles. The kit may be stored at 4°C for up to 6 months. For extended storage, it is recommended to store at -80°C.
