1. Prepare all reagents, samples and standards as instructed in the manual.
2. Add 50 µl of standard or sample to each well.
3. Add 50 µl of Bioaim mix.
4. Incubate 1 h at RT.
5. Wash and add 100 µl of TMB One-Step Substrate Reagent to each well.
6. Incubate 15 min at RT.
7. Add 50 µl of Stop Solution to each well.
8. Read at 450 nm/570 nm.
Platelet factor 4 (PF4) is a small cytokine belonging to the CXC chemokine family that is also known as chemokine ligand 4 (CXCL4). It is a 70-amino acid protein that is released from the alpha-granules of activated platelets and binds with high affinity to heparin. Its major physiologic role appears to be neutralization of heparin-like molecules on the endothelial surface of blood vessels, thereby inhibiting local antithrombin III activity and promoting coagulation. As a strong chemoattractant for neutrophils and fibroblasts by interacting with a splice variant of the chemokine receptor CXCR3, known as CXCR3B. PF4 probably has a role in inflammation and wound repair.
The heparin:PF4 complex is the antigen in heparin-induced thrombocytopenia, an idiosyncratic autoimmune reaction to the administration of the anticoagulant heparin. PF4 autoantibodies have also been found in patients with thrombosis and features resembling HIT but no prior administration of heparin.
The entire kit may be stored at -20°C for up to 1 year from the date of shipment. Avoid repeated freeze-thaw cycles. The kit may be stored at 4°C for up to 6 months. For extended storage, it is recommended to store at -80°C.
