1. Prepare all reagents, samples and standards as instructed in the manual.
2. Add 100 µl of standard or sample to each well.
3. Incubate 2.5 h at RT or O/N at 4°C.
4. Wash and add 100 µl of prepared biotin antibody to each well.
5. Incubate 1 h at RT.
6. Wash and add 100 µl of prepared Streptavidin solution to each well.
7. Incubate 45 min at RT.
8. Wash and add 100 µl of TMB One-Step Substrate Reagent to each well.
9. Incubate 30 min at RT.
10. Add 50 µl of Stop Solution to each well.
11. Read at 450 nm/570 nm.
Follicle-Stimulating Hormone (FSH) is a 33 kDa heterodimer belonging to the glycoprotein hormone family. It is composed of noncovalently linked glycosylated alpha and beta chains. The alpha subunit is also a component of Luteinizing Hormone (LH), Thyroid-Stimulating Hormone, and Chorionic Gonadotropin. The unique beta subunit confers the protein’s specific biological action and is responsible for the interaction with the FSH Receptor.
FSH is produced and secreted by the anterior pituitary gland. Its secretion is controlled by Gonadotropin-Releasing Hormone from the hypothalamus; however, FSH secretion can also be stimulated by activins and inhibited by inhibins. FSH works in concert with LH to regulate female reproduction; It stimulates follicular growth and LH induces ovulation. FSH can also regulate estrogen formation by inducing the aromatase enzyme that converts androgen to estradiol.
In males, FSH is critical for sperm production. Mutations in the human FSH and FSHR genes are associated with reduced reproductive functioning in females. Additionally, increased production of autoantibodies directed against FSH beta has been suggested to contribute to certain types of female infertility. Assisted reproductive technology commonly uses recombinant FSH to induce follicle growth.
The entire kit may be stored at -20°C for up to 1 year from the date of shipment. Avoid repeated freeze-thaw cycles. The kit may be stored at 4°C for up to 6 months. For extended storage, it is recommended to store at -80°C.
