1. Prepare all reagents, samples and standards as instructed in the manual.
2. Add 100 µl of standard or sample to each well.
3. Incubate 2.5 h at RT or O/N at 4°C.
4. Wash and add 100 µl of prepared biotin antibody to each well.
5. Incubate 1 h at RT.
6. Wash and add 100 µl of prepared Streptavidin solution to each well.
7. Incubate 45 min at RT.
8. Wash and add 100 µl of TMB One-Step Substrate Reagent to each well.
9. Incubate 30 min at RT.
10. Add 50 µl of Stop Solution to each well.
11. Read at 450 nm/570 nm.
CD200 antigen, also known as OX-2, is a type I transmembrane protein belonging to the Ig superfamily. The protein encoded by this gene is a type-1 membrane glycoprotein, which contains two immunoglobulin domains, and thus belongs to the immunoglobulin superfamily. Studies of the related genes in mouse and rat suggest that this gene may regulate myeloid cell activity and delivers an inhibitory signal for the macrophage lineage in diverse tissues. CD200 is widely expressed in multiple cell types. It interacts with CD200 R1, another Ig superfamily inhibitory receptor primarily expressed on leukocytes of the myeloid lineage, to inhibit myeloid functions. CD200 R1 is a transmembrane glycoprotein and member of the immunoglobulin superfamily. It is expressed primarily in cells of the myeloid lineage, while its ligand, CD200 R1 is though to propagate inhibitory signals and disruption of the CD200/CD200 R1 receptor-ligand pair leads to a predisposition to autoimmune phenotypes.
