• TMB One-Step Substrate
1. Prepare all reagents, samples and standards as instructed in the manual.
2. Add 50 µl of standard or sample to each well.
3. Add 50 µl of Bioaim mix.
4. Incubate 1 h at RT.
5. Wash and add 100 µl of TMB One-Step Substrate Reagent to each well.
6. Incubate 15 min at RT.
7. Add 50 µl of Stop Solution to each well.
8. Read at 450 nm/570 nm.
CXCL12 is expressed in many tissues in mice including brain, thymus, heart, lung, liver, kidney, spleen, and bone marrow. CXCL12 is strongly chemotactic for lymphocytes. During embryogenesis, it directs the migration of hematopoietic cells from fetal liver to bone marrow and the formation of large blood vessels. It has also been shown that CXCL12 signalling regulates the expression of CD20 on B cells. CXCL12 is also chemotactic for mesenchymal stem cells and is expressed in the area of inflammatory bone destruction, where it mediates their suppressive effect on osteoclastogenesis.
The entire kit may be stored at -20°C for up to 1 year from the date of shipment. Avoid repeated freeze-thaw cycles. The kit may be stored at 4°C for up to 6 months. For extended storage, it is recommended to store at -80°C.
