1. Prepare all reagents, samples and standards as instructed in the manual.
2. Add 100 µl of standard or sample to each well.
3. Incubate 2.5 h at RT or O/N at 4°C.
4. Wash and add 100 µl of prepared biotin antibody to each well.
5. Incubate 1 h at RT.
6. Wash and add 100 µl of prepared Streptavidin solution to each well.
7. Incubate 45 min at RT.
8. Wash and add 100 µl of TMB One-Step Substrate Reagent to each well.
9. Incubate 30 min at RT.
10. Add 50 µl of Stop Solution to each well.
11. Read at 450 nm/570 nm.
BAFF, also known as B-lymphocyte stimulator (BLyS), TNF- and APOL-related leukocyte expressed ligand 1 (TALL-1), and TNF homolog that activates apoptosis, NFKB and JNK (THANK), is a TNF ligand superfamily member and has been designated TNFSF13B. BAFF is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is expressed in B cell lineage cells, and acts as a potent B cell activator. It has been also shown to play an important role in the proliferation and differentiation of B cells. It is expressed as a membrane-bound type II transmembrane protein on various cell types including monocytes, dendritic cells and bone marrow stromal cells.
BAFF is the natural ligand of three unusual tumor necrosis factor receptors named BAFF-R (BR3), TACI and BCMA, all of which have differing binding affinities for it. BAFF-R is involved in the positive regulation during B cell development. TACI binds worst since its affinity is higher for a protein similar to BAFF, called a proliferation-inducing ligand (APRIL). BCMA displays an intermediate binding phenotype and will work with either BAFF or APRIL to varying degrees.
